Fig. 2A-C RNA blot analysis of MAT-1 and MAT-2 specific transcripts. Poly(A)+ RNAs (20 µg per lane) from MAT-1 strain C5 (lanes 1), MAT-2 strain C3 (lanes 2), or strains C5 plus C3 (lanes 1 + 2) grown in MM were analysed with the following probes. A MAT-1 idiomorph-specific PCR fragment #3 (Fig. 1). B MAT-2 idiomorph-specific PCR fragment #2 (Fig. 1). C Ethidium bromide- stained gel showing RNA size markers (lanes labeled RNA, sizes indicated on the right) compared with lambda DNA digested with HindIII (lane labeled k, sizes indicated on the left). The MAT-1 probe detects a 2.2± 0.1-kb transcript (A), the MAT-2 probe detects a 2.1±0.1-kb transcript (B). Both transcripts were detected in poly(A)+ RNA from a co-culture of strains of both mating types (A, B). No signals were found when MAT-1 RNA was probed with a MAT-2 specific probe and vice-versa. |
